James McClenahan is the son of John Burns McClenahan Sr. (1774–1847) and Lydia Burns (1774–1860).

He married to Matilda Manning on 10 Jan 1842 at Perryville, in Central Township, Perry County, Missouri. They had eight probable children:
- William McClanahan (1843–1850)
- Mary Elizabeth McClanahan (1844–1844)
- Michael Emmett McClanahan (1849–1854)
- Louis Joseph McClanahan (1849–1850)
- Cornelius McClanahan (1851–?)

James had a family disagreement with brother Madison, and changed the spelling of his surname to McClanahan. Over time, his descendants and many other family members also used this spelling.
