Sarah Jane Hamilton was born on October 10, 1849, in Red River County. She was the first-born child of her parents Rufus Hamilton and Martha Ballard Hamilton. She was the granddaughter of Robert Shaw Hamilton and his wife Elizabeth Bethea Hamilton. She was also the granddaughter of Barclay Moorman Ballard and Mary Gillentine Ballard.

Sarah did not marry. She and her younger sister Elizabeth lived in their parents' household until their deaths, helping their mother to care for their younger siblings. After her parents's deaths, Sarah and Elizabeth moved into the household of first their brother Robert and then their brother Davis.

Sarah died at the age of 82 on February 2, 1932, in Kaufman County, Texas, where she had lived most of her life. Her sister Mary Elizabeth who had been her life-long companion had died four months earlier. Sarah and Elizabeth were buried not far from their parents' graves in Radar Cemetery, Kaufman County, Texas.
