David was a son of William Meriwether (1730–1790) and Martha Cox (1732–1801). He was born and raised in Goochland County, Virginia, being baptized in Saint James Northam Parish there by Parson William Douglas on 5 Feb 1757.
About 1782 in Virginia David married his second cousin, Mary Lewis, daughter of John and Anne Lewis. David and Mary were the parents of six known children: Anne Lewis, Martha Wood, Jane, Henry Wood, Mary Lewis and Sarah Lewis Meriwether.
David died about 1795 in Jefferson County, Kentucky, at about the age of 41. His remains were removed from the original burial yard on the family farm to Cave Hill Cemetery in 1889.
